摘要
目的 探讨用双向电泳 (Twodimensionalelectrophoresis,2 DE)对大籽蒿花粉提取液蛋白质组分进行分析的可能性及其实用价值。方法 按常规方法提取大籽蒿花粉变应原 ,经SDS 聚丙烯酰胺凝胶电泳及双向电泳对大籽蒿花粉提取液变应原进行蛋白质组分分析。结果 单纯聚丙烯酰胺电泳结果可见有 4个较明显的条带 ,分子量分别为 6 2kD ,5 2kD ,4 3kD和 2 5kD。经双向电泳蛋白组分分析 ,发现大籽蒿花粉提取液有 85种蛋白组分。其中 ,第 139点的分子量为 31.4 9kD ,等电点为 5 .39;第14 3点的分子量为 31.86kD ,等电点为 6 .38。这两个点与文献所报道组分A2c的蛋白质分子量为31kD ,等电点分别为 5 .3和 6 .35一致。结论 双向电泳对大籽蒿花粉提取液蛋白质组分进行分析具有实用价值 。
Objective To study possibility of two dimensional electrophoresis (2 DE) in protein analysis of Artemisia allergen extract. Methods Artemisia allergen extract was prepared with routine method, SDS polyacrylamide gel electrophoresis (SDS PAGE),two dimentional electrophoresis (2 DE),laser imagescanning,2 DE gel analysis software were used to analyzing its protein components. Results There were four obvious bands in SDS PAGE, their molecular weights were 62kD,52kD,43kD and 25kD respectively. Artemisia pollen allergen had 85 protein components, whose molecular weights (MW) and isoelectric points (pI) were demonstrated in our table 2 in this paper. Our results had good repetition with our previous paper. Conclusions Two dimensional electrophoresis (2 DE) is a good method in protein analysis of Artemisia allergen extract, its potential value in allergic components needs to be further studied.
