摘要
目的研究目前文献中常用的计算方法和5-溴脱氧尿苷(BrdU)标记方法对不同生长状态肿瘤细胞潜在性倍增时间(Tpot)检测结果的影响。方法应用BrdU/DNA双参数流式细胞分析和相对运动法对鼻咽癌细胞株CNE进行研究。结果BrdU标记浓度10~100μmol/L、标记时间5~30min和相对运动时间1.5~8.5h对Tpot检测结果无显著影响(P<0.05);平台期CNE细胞未能被BrdU标记,而指数生长期CNE细胞和平台期CNE细胞换新鲜培养液后BrdU标记指数分别为48.8%和33.6%。结论本方法适宜于今后的临床应用,有助于解释临床上肿瘤加速再增殖发生的时间问题。
To improve control of measurement of potential doubling time for cancer cells,and to understand the influence of BrdU labeling concentration, time and length of relative movement as well as growth status of human nasopharyngeal carcinoma cell line CNE on such kinetic parameters as potential doubling time (Tpot).METHODS BrdU/DNA flow cytometry was used. Relative movement method(RM)was employed to calculate Tpot LI, etc.RESULTS (1)There were no statistical differences among the kinetic parameters for 10-100μmol/L BrdU labeling concentration with a 5 to 30 minutes of labeling time and 1. 5 to 8. 5 hours of relative movement; (2) BrdU labeling could not be detected in plateau phase CNE cells either before or after γ-ray irradiation,whereas is could be detected immediately after a change of fresh culture medium.CONCLUSIONS The method used here was evaluated with a view to clinical application. The influence of growth status of CNE cells on kinetic parameters might be related to 'delayed' accelerated repopulation observed in clinical fractionated radiotherapy.
