五味子多糖对lewis肺癌细胞株凋亡及FAS/FASL表达的影响 被引量:8

摘要 目的:探讨五味子多糖对lewis肺癌细胞增殖的影响,探究FAS/FASL在lewis肺癌细胞中的表达变化以及其对lewis肺癌细胞株凋亡的影响机制。方法:采用MTT法检测五味子多糖对lewis肺癌细胞增殖的抑制作用,收取细胞总蛋白并利用蛋白免疫印迹法检测其对FAS/FASL表达的影响。结果:五味子多糖能够抑制lewis肺癌细胞的增殖。五味子多糖能够促进lewis肺癌细胞FAS蛋白的表达,其中高剂量组为0.664+0.003,低剂量组为0.286+0.017,且两组间的差异具有统计学意义(P<0.05)。五味子多糖能够抑制lewis肺癌细胞FASL蛋白的表达,其中高剂量组为0.358+0.022,低剂量组为0.724+0.006,且两组间的差异具有统计学意义(P<0.05)。结论:五味子多糖能够抑制lewis肺癌细胞的增殖,其作用机制可能与调控Fas/FasL系统有关。 Objective To research the proliferation effect of Schizandra Chinensis Polysaccharide on Lewis lung cancer cell and detect the expression of FAS/FASL. Method Inhibitory effect of schizandra chinensis polysaccharide on the proliferation of Lewis lung cancer cells was detected by MTT. Total cellular protein was collected and the expression changes of FAS/FASL were detected by Western blot. Results Lewis lung cancer cell proliferation was inhibited by schizandra chinensis polysaccharide. The expression of FAS in high dose schizandra chinensis polysaccharide group was significantly higher than that in low dose schizandra chinensis polysaccharide group (0.664+0.003, 0.286+0.017, P<0. 05). The expression of FASL in high dose schizandra chinensis polysaccharide group was significantly lower than that in low dose schizandra chinensis polysaccharide group (0.358+0.022, 0.724+0.006, P<0.05). Conclusion The proliferation of Lewis lung cancer cells can be inhibited by schizandra chinensis polysaccharide and the mechanism may be related to influencing the expression of FAS/FASL.
