四川省猪繁殖与呼吸综合征病毒分子流行病学调查 被引量:6

Molecular epidemiological investigation of porcine reproductive and respiratory syndrome virus in Sichuan province

摘要 为了解2012-2013年四川省猪繁殖与呼吸综合征病毒(PRRSV)流行毒株的遗传变异背景和分子流行病学情况,对PRRSV阳性病料中的Nsp2、ORF5基因进行RT-PCR扩增和序列测定,所获得的序列分别与美洲型代表株(VR2332)、欧洲型代表株(LV)、标准毒株和变异毒株进行相似性分析。结果本研究成功获得8条Nsp2基因序列和17条ORF5基因序列,序列分析表明所有PRRSV流行毒株均属于美洲型,其中8株Nsp2基因序列与标准毒株和变异毒株的核苷酸相似性分别为75.2%~88.6%和94.8%~97.1%,推导的氨基酸相似性分别为58.7%~79.2%和92.8%~94.8%;17株ORF5基因序列与标准毒株和变异毒株的核苷酸相似性分别为90.1%~96.6%和95.6%~99.4%,推导的氨基酸相似性为77.2%~93.0%和88.0%~96.8%;与标准毒株相比,8株Nsp2基因有30个不连续氨基酸缺失,17株ORF5基因序列有点突变。从遗传进化分析看,所有流行毒株均属于PRRSV亚群Ⅲ。 To understand the genetic variation and molecular epidemiology of porcine reproductive and respiratory syndrome virus( PRRSV) in Sichuan province during 2012-2013. Nsp2 and ORF5 genes were amplified by RT-PCR and sequenced for PRRSV isolates. The sequences were genetically compared with American representative( VR2332),European representative( LV),Chinese standard and mutated strains. Eight Nsp2 and 17 ORF5 genes were acquired in this study. Sequence analysis suggests that all isolates belong to American type. The 8 Nsp2 genes share about 75. 2%-88. 6% and 94. 8%-97. 1% identity in nucleotide sequences,and 58. 7%-79. 2% and 92. 8%-94. 8% identity in deduced amino acid sequences,with the standard strains and mutants,respectively. The 17 ORF5 genes share about 90. 1%-96.6% and 95. 6%-99. 4% identity in nucleotide sequences,and 72. 3%-93. 0% and 88. 0%-96. 8% identity in deduced amino acid sequences,with the standard strains and mutants,respectively. Compared to the standard strains,the 8 Nsp2 genes have the continuous deletion of 30 amino acids,and the 17 ORF5 genes have single point mutations. All PRRSV isolates belong to subgroup III from phylogenetic analysis in this study.
