鸭源H5N1亚型禽流感病毒的分离鉴定及其HA基因和NA基因的序列分析 被引量:3

摘要 本研究旨在了解鸭源H5N1亚型禽流感病毒的流行及遗传进化特点。采集健康鸭的肛门拭子,常规处理后接种于10日龄SPF鸡胚,收取24~96h内死亡鸡胚尿囊液,进行血凝和血凝抑制试验、核酸鉴定及HA和NA基因序列分析。结果表明,该分离株为禽流感病毒H5N1亚型,命名为A/Duck/Liaoning/N/2011(H5N1)。进化分析显示,HA基因的开放阅读框全长1 704nt,编码567个氨基酸;NA基因的开放阅读框全长1 350nt,编码449个氨基酸。HA蛋白裂解位点的氨基酸序列为PQRERRRKR,含有多个碱性氨基酸,具有高致病性禽流感病毒的分子特征。该毒株属于新型H5N1亚型禽流感病毒2.3.2.1系,主要在东亚地区流行。该研究结果为进一步研究鸭源H5N1亚型禽流感病毒的分子进化提供了重要信息。 The study aimed to study the epidemic and genetic evolution characteristics of H5N1 subtype avian influenza virus isolated from ducks.Anal swabs of healthy duck were collected,and then were inoculated into 10-day-old SPF chicken embryos after conventional treatment.Allantoic fluid was collected from chicken embryos died within 24 to 96h.Hemagglutination assay and hemagglutination inhibition test,the RT-PCR identification,and nucleotide sequence analysis of HA and HN genes were conducted.The results showed that the isolate was identified as an H5N1 subtype avian influenza virus and named as A/Duck/Liaoning/N/2011(H5N1).Evolution analysis indicated that the open reading frame length of HA gene was 1 704 nt and encoded 567 amino acids,and the open reading frame length of NA gene was 1 350 nt,encoding 449 amino acids.The amino acid sequence of cleavage site in HA protein was PQRERRRKR,it contained multiple basic amino acid,that has the unique molecular characteristics of highly pathogenic avian influenza virus.The strain belonged to clade 2.3.2.1of a new H5N1 subtype avian influenza virus,which prevalent in eastern Asia.The study result provided important information for the further study of molecular evolution on H5N1 subtype avian influenza virus from ducks.
