摘要
目的通过研究胰岛素样生长因子-Ⅰ(IGF-Ⅰ)对骨骼肌运动损伤后卫星细胞再生的影响,探讨IGF-Ⅰ在骨骼肌运动损伤修复中的应用前景。方法使用SD大鼠1周力竭模型,首次力竭后第1、3、5天给予右侧腓肠肌中部的肌膜下分别注射外源性IGF-Ⅰ300ng/ml(实验组)、生理盐水1ml(对照组),分别在首次力竭后7、9、11、17、24、30d取材,进行骨骼肌HE染色光镜观察,电镜超微结构观察和增殖细胞核抗原(PCNA)表达的比较。结果首次力竭后7、11、17d实验组卫星细胞明显增多,对照组胶原纤维增多,30d实验组肌纤维塑形良好,对照组可见瘢痕愈合;电镜观察对照组可见肌浆网扩张,线粒体肿胀,还有凋亡细胞出现;实验组线粒体形态较正常,糖原颗粒增多,可见新生的肌纤维。PCNA指数实验组明显高于对照组(P<0.05)。结论IGF-Ⅰ对运动性骨骼肌损伤后的卫星细胞再生有促进作用。
Objective To study the effect of insulin-like growth factor I (IGF-Ⅰ) on the regeneration of satellite cells after sports injury, and to explore the application of IGF-Ⅰ in repair of skeletal muscle after sports injury. Methods The SD rat model of 1 week exhaustion was applied. On the 1st, 3rd, and 5th days after the first exhaustion, IGF-Ⅰ of 300ng/1ml was injected into the middle of right gastrocnemius muscle in the treatment group. The control group was injected with 1ml of saline. The samples were taken on the 7, 9, 11, 17, 24, and 30th days after the first exhaustion. The skeletal muscle was dyed with HE and observed with optical microscope and electric microscope. The expressions of proliferating cell nuclear antigen (PCNA) in it were compared. Results On the 7, 11, and 17th days after the first exhaustion, the satellite cells decreased significantly in the treatment group, but the collagen fibers increased in the control groups. On the 30th day, the muscle fibers were in good shape in the treatment groups, but the injured muscle healed with scar in the control group. Observation by electric microscope showed that the shape of biolast was normal and new born muscle fiber could be found in the treatment groups, while expanded sarcoplasmic reticulum,increased glycogen granules, engorged biolasts and apoptotic cells could be found in the control groups. The PCNA exponent in the treatment group was obviously higher than that in the control group. Conclusion IGF-Ⅰ can accelerate the regeneration of skeletal muscle satellite cells after sports injury.
