摘要
利用人单核细胞表面有纤维连接蛋白受体的特点,我们建立了一种分离、纯化人外周血单核细胞的方法,该法制备的细胞悬液单核细胞占97%以上,纯度明显优于Ficoll-Hypaque密度梯度离心法。单核细胞获得率近5倍于传统粘附法,且细胞活性保持在97%以上,是一种分离单核细胞的简单而有效的方法。
A simple and efficient method of monocyte seperation was developed based on the fact that fibronectin receptors are present on the membrane of human monocytes. The percentage of mono-cytes was beyond 97% in cells obtained by this method, it was significantly higher than that obtained by Ficoll-Hypaque centrifugation. The yield of monocytes was 5 times more than that with classical adherance method, and the cell viability was excellent(>97%). We concluded that this method is a simple and useful technique in seperating human peripheral blood monocytes.
