摘要 目的:建立高效液相色谱法测定酪丝亮肽中醋酸的含量,探讨化学合成多肽中醋酸的含量测定方法。方法:采用C18色谱柱(5μm,4.6 mm ×250 mm);流动相A为0.2 mol·L-1硫酸盐缓冲液(pH 2.3),B为乙腈-水(90:10);流速1.0 mL·min-1;先用流动相A-B(96:4)等梯度洗脱出醋酸峰至5 min,继用流动相A、B梯度洗脱多肽等其他强保留组分;紫外检测波长:220 nm;进样量:20μL。结果:方法的线性范围为1.0-5000μg·mL-1(r=0.9997);最低检测限20 ng;精密度RSD为0.83%(n=7);平均回收率为99.07%,RSD为0.49%(n=9)。结论:方法简便灵敏,结果准确可靠,可用于酪丝亮肽及其他合成多肽中醋酸的含量测定。 Objective:To establish a RP-HPLC method to determine the content of acetic acid in tyroserleutide and other synthetic peptides. Method:The HPLC system consists of C18 column(5μm,4. 6 mm ×250 mm) ,0. 2 mol·L-1 sodium sulfate buffer solution(pH 2. 3) as mobile phase A and acetonitrile - water (90=10) as mobile phase B with UV detection at 220 nm. First mobile phase A -B (96=4) is used to elute acetic acid for 5 minute, then A and B are used as mobile phase for the gradient elution of peptide and other strongly retained substances at a flow rate of 1.0 mL·min-1 ,the injection volume is 20 μL. Results:The linear range of the method is 1. 0 -5000 μg·mL -1 (r=0.9997); the limit of detection is 20 ng; the precision RSD is 0. 83% (n =7); the average recovery is 99. 07% ,RSD is 0.49% (n=9). Conclusion:The method is simple,sensitive,accurate and can be used for the determination of acetic acid in tyroserleutide and other synthetic peptides.
