摘要
目的 从日本血吸虫 (Schistosomajaponicum ,Sj)成虫cDNA文库中获得并分析日本血吸虫新的蛋白基因 ,为日本血吸虫病的防治提供侯选疫苗和药物靶点。方法 构建日本血吸虫成虫cDNA文库 ,挑取重组阳性克隆进行测序 ,对部分序列进行步移法测序获取全长cDNA ,并进行生物信息学分析和登录。结果 获得了 1个日本血吸虫新基因 -钙调神经磷酸酶 (Calcineurin ,CaN)基因B(AY2 2 0 75 1) ,长 12 6 6bp ,编码 16 9个氨基酸 ,含有 2个“EF -hand”Ca+2 结合区域 ,与曼氏血吸虫CalcineurinB有 84 %的同源性。
A new gene from adult worms of Schistosoma japonicum,calcineurin B gene is reported in this study.The cDNA library was constructed from adult worms of S.japonicum (Chinese strain).Clones selected randomly from the cDNA library were sequenced and the new gene was analyzed in GenBank database by BLAST programs and other programs.The calcineurin B gene with 1266 bp and coding for 169 amino acids was thereby obtained,and this new gene was submitted to GenBank as AY220751.The calcineurin B gene of S.japonicum had two “EF hand” calcium binding regions,and showed 84% of homology with the calcineurin B gene of S.mansonii.It proves that combination of the primer walking sequencing and bioinformatics technique is a feasible method to discover new genes in S japonicum
