摘要
目的 观察EB病毒 (Epstein Barrvirus,EBV)及其膜表面糖蛋白 35 0 (gp35 0 )对分离的脐带血CD5 + B细胞产生IL 6和免疫球蛋白的影响。方法 无菌采集新生儿脐带血 ,RosetteSepTM法分离总B细胞 ,免疫磁珠分离CD5 + B细胞 ,用EBV及纯化的gp35 0刺激细胞 ,ELISA法检测培养上清液中的IL 6和免疫球蛋白IgG、IgM ,对照为未加刺激的培养细胞上清液。结果 CD5 + B细胞于刺激的第 2 4h开始分泌IL 6 ,OD值较对照组显著升高 (P <0 .0 1) ;EBV刺激的CD5 + B细胞 13d以后的培养上清液中能检测到免疫球蛋白IgM和抗角蛋白自身抗体 (AKautoAb)IgM ;gp35 0刺激的CD5 + B细胞存活 15d ,其第 13、15天的细胞培养上清液中能检测到免疫球蛋白IgM ;未刺激的细胞存活 7d ,培养上清液中未检测到IL 6和免疫球蛋白。结论 EBV可能通过其膜表面糖蛋白gp35 0刺激培养的CD5 + B细胞产生IL 6和具有天然自身抗体性质的免疫球蛋白。
Objective To explore the role of Epstein-Barr virus and it's glycoprotein-350 in production of cytokine and immunoglobins by cultured CD5 +B cells. Methods Blood of umbilical cord was collected in sterile condition. B cells were isolated by the method of RosetteSep TM. CD5 + B cells were obtained using mouse IgG magnetic beads. These CD5 + B cells were stimulated by Epstein-Barr virus and it's gp350, respectively. ELISA was used to detect the supernatant. IL-6, IgG,and IgM in the unstimulated cells were used as negative control. Results We can detect IL-6 in supernatant of these cells after 24 h , the OD values increased significantly as compared with that of the con- trol (P<0.01). OD values of supernatant IgM and AK auto Ab IgM of the cells stimulated by Epstein-Barr virus increased significantly after 13 d. The cells stimulated by gp350 survived 15 d. OD values of IgM in the supernatant of the 13th and the 15th day culture were increased . The unstimulated cells survived 7 d. Conclusion These results suggest that Epstein-Barr virus can stimulate cultured CD5 +B cells to produce IL-6 and immunoglobulin which may possess the characteristics of natural auto-antibody by its glycoprotein-350.
