基于原子力显微镜抬高模式的DNA分子高度测量 被引量:3

摘要 在传统的轻敲模式原子力显微镜基础上利用抬高模式对DNA分子的高度进行了测量研究. 通过不断抬高针尖高度, 逐步减小针尖对样品的作用力, 以测量软样品的形变量, 并通过记录针尖的抬高高度来计算DNA分子的高度. 实验中利用抬高模式测得的DNA分子高度为1.5±0.2 nm, 而传统轻敲模式下测得的DNA分子的高度为0.8±0.2 nm, 表明针尖对DNA的作用力是导致传统轻敲模式测得DNA分子高度较低的重要因素. 同时本文采取的方法也适用于测量其他软样品的高度.
