卵黄抗体对ETEC粘附仔猪小肠上皮细胞的体外抑制作用研究 被引量:6

Adhesive inhibition of ETEC to piglets intestinal epithelial cells by egg-yolk antibodies against ETEC fimbriae in vitro

摘要 采用热抽提法提取 4种肠毒素性大肠杆菌菌毛蛋白 :K88、K99、F41和 987p。分别制成单价或多价的菌毛蛋白白油佐剂抗原 ,对产蛋鸡进行胸部肌肉分点注射免疫 ,初免后 2周加强免疫 1次。收集高效价卵黄抗体。用所获得各卵黄抗体对体外分离的初生仔猪小肠上皮细胞进行体外粘附抑制试验。结果表明 ,各种菌毛卵黄抗体均能特异地显著抑制相应大肠杆菌对仔猪上皮细胞的粘附 。 Four kinds of enterotoxigenic Escherichia coli (ETEC) fimbriae:K88,K99,F41 and 987p were extracted with thermoextract method. Vaccines were made from each fimbriae antigen, the mixture of four fimbriae antigen and white oil adjuvant. Laying hens were immunized with different vaccines. Two weeks later, one booster injection was given. The titer of egg-yolk antibodies was detected with micro-agglutination. Egg-yolk antibodies were collected and stored in 4℃. The results by tests of adhesive inhibition in vitro indicated that the adhesion of bacterial cells to intestinal epithelial cells was strongly inhibited by homologous anti-fimbrial antibody and the multi-valuent antibodies against fimbriae significantly decreased the number of any kinds of ETEC cells to the intestinal epithelial cells.
