摘要
目的 将活血通腑方药剂用于腹腔粘连大鼠 ,通过血浆纤维蛋白原 (FIB)含量、TGF βmRNA表达的检测和大鼠腹腔粘连级别评分 ,确立活血通腑方有效剂量。方法 将模型SD(清洁级 )雄性大鼠随机分为大、中、小剂量 3组 ,进行胃内药物灌注 ,观察d 7大鼠腹腔粘连情况 ,测定血浆FIB含量、粘连组织标本TGF βmRNA表达 ,筛选出活血通腑方的有效剂量。结果 ①血浆FIB的析出中、小、大剂量组呈递增态势 ;②粘连级别中、小、大剂量组顺序递增 ;③TGF βmRNA表达大、中、小剂量组递减。结论 中剂量活血通腑方能影响大鼠术后腹腔粘连程度、降低血浆FIB含量及TGF βmRNA表达。
OBJECTIVE To establish effective dosages for the prescription for activating blood circulation and removing obstruction in the fu-organs. METHOD Model SD male rats were divided randomly into large, medium and small dosage groups and were treated respectively with the prescription. Observation was made on the abdominal adhesion, the FIB content and the expression of the adhesive tissue sample TGF-βmRNA, in order to find the effective dosage for the prescription. RESULT ①FIB lixiviation was obvious in the medium dosage group and more obvious in the small dosage group (P<0.05~0.01);②adhesion grade increase progressively according to medium,small and large dosage group. ③The expression of TGF-βmRNA was degressive in the large, medium and small groups;CONCLUSION Medium dosage of the prescription can affect the abdominal adhesion and reduce FIB content and TGF-βmRNA expression of post-operative rats.
