摘要
为了研究细粒棘球绦虫EgA31重组蛋白的抗原性 ,作者将细粒棘球绦虫 6 6kDa抗原的cDNA克隆EgA31 3′端 5 0 0bp碱基片断亚克隆入pGEX 5X表达质粒 ,构建EgA31 GST重组蛋白原核表达系统 ,所制备的重组蛋白免疫豚鼠获得抗血清 ;免疫试验表明该抗血清可识别EgA31 GST重组蛋白及细粒棘球绦虫 6 6kDa抗原 ,也可与多房棘球蚴、犬复孔绦虫、犬钩虫、日本血吸虫的成虫虫体总蛋白 6 6kDa抗原分子发生交叉反应 。
In order to study the antigenicity of EgA31 recombinant protein derived from Echinococcus granulosus, a fragment of 500 bp from EgA31 3′ cDNA was subcloned into the expression vector pGEX 5X.EgA31/GST fusion protein was expressed in E.coli and purified.An antiserum was prepared by immunizing a Guinea pig with this fusion protein.The antiserum recognized the fusion protein and the 66 kDa antigen of E.granulosus ,and also cross reacted in Western blot with the 66 kDa antigenic molecule from other parasitic helminth species,including E.multilocularis,Dipylidium caninum,ancylostoma caninum,Schistosoma japonica, suggesting that this protein be a important factor for the survival of these helminthic parasites.
