摘要
以桃幼胚作为根癌农杆菌介导转化的受体,通过对GUS基因瞬时表达率的分析,研究此转化体系的最佳实验参数。实验结果表明,预培养时间、感染时间、共培养时间和根癌农杆菌诱导物AS等对转化效率都有一定的影响。预培养1d、感染15min、共培养42h和不添加AS时GUS瞬时表达率最高。
Using transient expression of GUS gene,the optimal conditions for Agrobacterium-mediated transformation of peach immature embryos was investigated. Histochemical GUS assay showed that the transient expression rate was infected by pre-culture time, infection time, co-culture time and the way of adding acetosyringone. Pre-culture 1 d, infection 10 to 15 min, co-culture 42 h and no add of acetosyringone contributed to the highest level of transient expression.
