在KC,VW和MS三种基本培养基中,MS最适宜万带兰种子的萌发。外加香蕉汁100 g/L可明显提高种子发芽率并促进幼苗生长。 MS加BA(0.5~1 mg/L)和NAA(0.05 mg/L)可诱导万带兰种子在膨大后产生愈伤组织,在试验条件下,BA用量在0.5 mg/L以上时,与愈伤组织产生呈正相关,而与种子发芽率却呈负相关。MS加BA0.3 mg/L,NAA0.05 mg/L有利于万带兰幼苗的生长,同时酪蛋白水解物和香蕉汁均对生长有促进作用。活性炭可有效防止幼苗变褐死亡。未见NaOH处理万带兰种子和黑暗条件能影响其萌发,但播种密度对种子萌发有影响,呈现"群体效应"。
MS medium was the best for the germination of Vanda seeds among 3 media, KC, VW and MS. The germination rate was increased and the seedling growth was promoted by adding 100 g/L of banana juice. MS medium adding BA (0.5—1.0 mg/L) and NAA (0.05 mg/L) induced the expansion of Vanda seeds which produced calli then. Under the experimental conditions, when 0.5 mg/L or higher concentrations of BA were added, the added rate of BA was positively correlated with the production of calli but negatively correlated with the germination rate of seeds. MS medium adding 0.3 mg/L of BA and 0.05 mg/L of NAA favoured the growth of Vanda seedlings. Protein hydrolysates and banana juice also promoted the seedling growth. Active carbon were effective for prevent the seedlings from browning and dying. Neither NaOH treatment nor dark culture showed any significant effects on the seed germination. A group effect was observed in Vanda through the seed sowing density experiments.
