摘要
目的 检测翼状胬肉组织中血管生长因子VEGF、FGF 2和抑制因子PEDF的表达 ,以研究PEDF在翼状胬肉发展中所起作用。方法 用免疫荧光组化法及免疫印迹法对 9例原发性翼状胬肉组织进行PEDF、VEGF及FGF 2的检测、分析 ,并与 8例正常人结膜、角膜组织进行对照研究。结果 PEDF在正常人结膜、角膜上皮中高表达 ,而在翼状胬肉组织中没有或弱表达。VEGF和FGF 2在翼状胬肉组织中高表达 ,而在正常人结膜、角膜组织中表达较弱。结论 在翼状胬肉组织中血管生长抑制因子PEDF没有或弱表达 ,而血管生长因子VEGF和FGF 2高表达 。
Objective To compare the expression levels of pigment epithelium-derived factor(PEDF) and vascular endothelial growth factor(VEGF) and fibroblast growth factor-2(FGF-2)in pterygia. Methods The normal human conjunctival and corneal tissues (8 cases) and pterygial tissues (9 cases) were collected for immunofluorescence study using monoclonal antibodies specific to human PEDF and VEGF and a polyclonal antibody specific to human FGF-2.PEDF and VEGF were measured by Western blot analysis. Results High levels of PEDF immunostaining were detected in all of the 8 independent normal human conjunctival and corneal specimens.PEDF was predominantly expressed in the epithelium of the conjunctiva and cornea.Under the same immunostaining conditions,however,PEDF was not detected in the epithelium or the fibroangiogenic mass of all of the 9 pterygia.In contrast,VEGF and FGF-2 were both detected in pterygia tissues,especially surrounding the blood vessels.Average PEDF level in pterygia was drastically lower than those in normal cornea and conjunctiva,respectively,but that of VEGF was significantly higher. Conclusion Normal human conjunctiva and cornea express PEDF at high level,and pterygium expresses FGF-2 and VEGF but lacks PEDF expression.This imbalance between angiogenic stimulators and inhibitors may play an important role in the formation and progression of pterygium.
