摘要
猪的体细胞核移植研究在生物医学领域很有意义。本试验对克隆技术中关键步骤之一的去核方法作了深入研究。结果表明,由本实验室改进的Willadsen去核法--二步挤压去核法无论在去核操作成功率(73.9%)上,还是在去核效率(81.2%)上都显著地高于McGrath-Solter去核法(42.5%、67.4%,P<0.05)。还发现,卵母细胞成熟培养36 h后去核组去核效率(89.1%)显著地高于成熟培养44 h后去核组(55.8%,P<0.05)。
The study on porcine somatic nuclear transfer is of the great meaning in the field of biomedicine. This experiment was conducted to study enucleated method, which is one of the key techniques for animal cloning. The results show that the modified Willadsen's method for enucleation, called two-step-squeezing method, is significantly better than McGrath-Solter's method in the enucleated rates (73.9%, 42.5%, P<0.05) and the rates (81.2%, 67.4%, P<0.05) of absolutely removing nuclei matter. It was also found that the rates (89.1%) of absolutely removing nuclei matter for 36h-matured-in-vitro oocytes is significantly higher than that for 44h group (55.8%, P<0.05).
