摘要
目的:建立反相高效液相色谱法测定弱威氏霜中苯甲酸和水杨酸的含量.方法:采用C18色谱柱(39 mm×150 mm,5μm),流动相为甲醇:0.02 mol/L磷酸二氢钠溶液(磷酸调pH=5.0)(55:45),流速1.0 m1/rmin,检测波长225nm.结果:苯甲酸和水杨酸分别在8.0~40.0μg/ml(r=0.9999)和4.0~20.0 μg/ml(r=0.9998)范围内线性关系良好,低、中、高浓度回收率为99.38%~101.01%(RSD 为0.48%~0.98%).测定三批样品,含量合格.结论:该方法简便、快速、测定结果准确,符合质量控制要求,适合于该制剂的含量测定.
Objective: To establish a RP-HPLC method for determination of benzoic acid and salicylic acid in cream Whitfield's (Weak). Methods: The chromatographic conditions were as follows: C18 column (39 mm×l50 mm, 5 μm), methanol and 0.02 mol/L NaH2PO4 solution (pH adjusted to 5.0 or so with H3PO4) (55∶45)were used as mobile phase, at a flow rate of l.0 ml/min and the detection wavelength of 225 nm. Results: The linear range of calibration curve of benzoic acid and salicylic acid was 8.0 ~ 40.0 μg/ml (r=0.9999). 4.0 ~ 20.0 μg/ml (r=0.9998) respectively. The recoveries of low, medium and high concentration were within 99.38% ~ 101.01% (RSD=0.48% ~ 0.98%). Three samples were analysed and the results were satisfied. Conclusion: This method is simple, rapid and accurate, and can be used for the quality control of the preparation, and is suitable for determination.
