高效液相色谱荧光法测定人血中去甲长春花碱的浓度 被引量:2

摘要 目的 :建立测定血浆及全血中去甲长春花碱 (VNB)浓度的HPLC方法。方法 :采用Hypersil C1 8分析柱 ,甲醇 0 .0 5mol·L-1 磷酸二氢钾缓冲液 (pH 4 .0 ) 四氢呋喃 (45∶5 2 .5∶2 .5 )为流动相 ,流速 1 .0mL·min-1 ,荧光检测波长Ex =2 80nm ,Em=36 0nm ,进样 5 0 μL。 结果 :VNB血浆标准曲线方程为 :C =2 .76× 1 0 -4A +0 .80 2 5 (r =0 .9994 ) ,日内RSD为 5 .6 7%,日间RSD为 7.2 9%,方法回收率为 (1 0 1 .4± 5 .7) %;VNB全血标准曲线方程为 :C =5 .34× 1 0 -4A +0 .4 384 (r =0 .9998) ,日内RSD为 5 .99%,日间RSD为 7.2 3%,方法回收率为 (1 0 0 .0± 6 .0 ) %。线性范围均为 1~ 1 0 0 0 μg·L-1 。结论 :本方法灵敏、准确 ,能够应用于临床药学研究。 OBJECTIVE To establish a method for determination of vinorelbine in human plasma and blood by HPLC.METHODS 1 mL plasma or 0.5 mL blood was extracted by diethyl ether and re-extracted by phosphate buffer,and 50 μL buffer was injected. A Hypersil-C 18 column was used and the mobile phase was composed of methanol-buffer of KH 2PO 4 (pH 4.0)-tetrahydrofuran (45∶ 52.5∶ 2.5). Flow rate was 1.0 mL·min -1. A fluorescence detector (excitation wavelength: 280 nm; emission wavelength: 360 nm) was used.RESULTS The calibration curve of equation VNB in plasma was C= 2.76×10 -4A+ 0.802 5 (r= 0.999 4), RSD of intra-day and inter-day tests was 5.67% and 7.29%, respectively. The relative recoveries of the methods with plasma and blood sample were( 101.4± 5.7)% and ( 100.0± 6.0)%, respectively. While those of VNB in blood was C= 5.34×10 -4A+ 0.438 4 (r= 0.999 8), 5.99% and 7.23%, respectively. Both liner ranges were 1~ 1 000 μg·L -1. CONCLUSION The established assay is sensitive and accurate, which can be applied to clinical pharmacy research.
