摘要
目的 建立全自动测定腺苷脱氨酶 (ADA)的酶偶联分光光度法。方法 用酶联比色法对pH值、腺苷浓度等反应条件及各种实验参数进行研究。用该法测定了 90例健康者的血清样本及 99例胸腹水的ADA活性。结果 该法最低检出限为1.7U/L ,线性范围可达 2 2 0U/L(r=0 .9989) ,批内CV为 2 9%~ 5 .4 % ,批间CV为 4 .7%~ 6 4 % ,回收率为 99.7%。胆红素 <136 .8μmol/L ,血红蛋白 <3.5g/L ,抗坏血酸 <2 84 μmol/L和三酰甘油 <9.0 3mmol/L的情况下 ,对测定结果无显著性影响。 结论 本法灵敏度高 ,线性、精密度好 ,结果准确 ,干扰少 ,且操作简便快速 ,适用于全自动生化分析仪分析。
Objective To establish an automated enzyme-coupled spectrophotometric assay for determination of adenosine desaminase (ADA).Methods The reaction conditions,e.g.,pH,substrate concentration and so on were optimized,and the parameters in the assay were set up.The serum levels of health individual (n=90) and ADA levels in the pleural fluid or ascites (n=99) were determined and the assay was evaluated.Results The calibration curve for adenosine desaminase was linear between 1.7U/L and 220U/L.The inter and intra CV were 5.35% and 6.38% respectively.The mean recovery rate was 99.7%. This method was free from interference by brilirubin (<136.8 μmol/L),hemoglobin (<3.5g/L),ascorbate (<284 μmol/L) and triglycerides (<9.03mmol/L).Conclusion The sensitivity,linearity and precisions of the established assay were satisfied without noticeable interference.The procedure was accurate,rapid and automatable to be used in clinical assay.
