重组人肝再生增强因子抑制大鼠肝星状细胞c-jun基因表达的研究 被引量:10

摘要 目的探讨重组人肝再生增强因子(hALR)对大鼠肝星状细胞c-jun基因表达的影响。方法在培养的肝星状细胞系中加入200、20、2 μg/L三种浓度的hALR,于8、24、48、72 h四个时间点收集细胞,提取总RNA;用逆转录-聚合酶链反应(RT-PCR)方法测定c-jun的基因表达水平。结果对照组肝星状细胞有c-jun的明显表达;不同浓度的hALR 3组肝星状细胞c-jun基因表达水平在8、24、48、72 h四个时间点均明显低于对照组;2μg/L组为对照组的60%-64%,20μg/L组为对照组的41%-43%,200 μg/L组为对照组的29%-35%。与对照组比较差异有显著性(P<0.01)。结论hALR对大鼠肝星状细胞c-jun基因表达有明显的抑制作用。 Objective To investigate the effects of recombinant human augmentor of liver regeneration (hALR) on c-jun gene expression in hepatic stellate cell. Methods Hepatic stellate cell (HSC-T) was cultured in medium containing different concentrations of hALR (200 μg/L, 20 μg/L, 2 μg/L respectively) and cells were collected at different incubation period (8, 24, 48, 72 h). Total RNA of HSC was isolated and c-jun gene expression levels were measured by reverse transcription polymerase chain reaction. Results c-jun gene expression levels of HSC in high (200 μg/L), middle (20 μg/L) and low (2 μg/L) concentrations of 3 hALR groups were much lower than those in control group after exposure to hALR at 8,24,48 and 72 h. Low dose group was 60-64% of control group, middle dose group was 41-43% of control group and high dose group was 29-35% of control group. Conclusions hALR could inhibit the gene expression of c-jun in hepatic stellate cell.
