大鼠骨基质形态增龄性改变及其与骨代谢指标的相关性研究 被引量:3

Age-related changes of rat bone histomorphometry and its correlation to other parameters of bone metabolism

摘要 目的 研究大鼠增龄过程中骨基质形态计量改变规律及与骨代谢指标相关性。 方法  3、9、15个月龄雌性SD大鼠各 10只。第 4腰椎及右侧胫骨上段作形态计量学分析。物理密度法测定股骨及腰椎骨密度 (BMD)。第 3腰椎作抗压缩试验。自动分析仪测定血清钙、磷 ,放免法测定1,2 5 (OH) 2 D3 。 结果 与 3月龄组比较 ,15月龄组第 4腰椎骨小梁体积减小 4 3 3% (P <0 0 1) ,胫骨上段骨小梁体积减小 2 8 0 % (P <0 0 1) ,骨形成表面降低 17 2 % (P <0 0 5 ) ,骨吸收表面增加39 8% (P <0 0 1) ,腰椎、股骨BMD分别下降 7 7% (P <0 0 1)和 7 3% (P <0 0 1)。骨小梁体积与腰椎BMD、最大载荷、血清 1,2 5 (OH) 2 D3 呈明显正相关 (r =0 6 84、0 972及 0 86 6 ,P <0 0 0 1)。血清 1,2 5 (OH) 2 D3 与胫骨骨形成表面及矿化沉积率呈明显正相关 (r =0 5 87,r =0 6 2 8,P <0 0 1)。 结论  15月龄雌性SD大鼠骨形成减弱 ,骨吸收增加 ,骨量减少 ,骨结构改变 ,生物力学性能降低。骨形态计量学指标与腰椎BMD和骨生物力学指标有明显的相关性。 Objective To study the age-related changes of rat bone histomorphometry and its correlation to other parameters of bone metabolism. Methods Ten female SD rats at the age of 3 months old,9 months old and 15 months old were labeled with tetracycline(30 mg/kg) via abdomen on fourteen and four days before executing. Histomorphometry were performed on L4 and right tibiae. Physical density of left femur and L2 were measured. Anti-compression property of L3 was assayed. Serum levels of calcium and phosphorus and 1,25(OH)_ 2 D_ 3 were determined. Results Compared with the 3-month-old rats,the trabecular bone volume(TBV) of L4 and right tibiae decreased 43.3%( P <0.01)and 28.0%( P <0.01),respectively in 15-month-old rats;Formation surfaces(FS) of lumbar and femur decreased 17.2%( P <0.05),Resorption surfaces(RS) increased 39.8%( P <0.01),Bone density(BMD) decreased 7.7% ( P <0.01) and 7.3%( P <0.01),respectively. Compared with the 3-month-old group,maximal load,maximal stress and modeling volume respectively decreased 53.8%( P <0.01),53.7%( P <0.01)and 33.2%( P <0.01) in 15-month-old group. Serum 1,25(OH)_ 2 D_ 3 declined 28.6%( P <0.05)in 9-month-old group and 34.1%( P <0.01)in 15-month-old group. A positive correlation was found between TBV and lumbar vertebrae BMD( r =0.684,P <0.001),serum 1,25(OH)_ 2 D_ 3 (r=0.866,P <0.001),maximal load( r =0.972,P <0.001),maximal stress( r =0.971,P <0.001) and modeling volume( r =0.971,P <0.001). There were also a positive correlation between serum 1,25(OH)_ 2 D_ 3 and FS( r =0.587, P <0.01) and mineral apposition rate(MAR)( r =0.628, P <0.01). Conclusions The bone formation was decreased while its resorption was increased;the bone mass was declined and its micro-structure was changed in fifteen-months-old rats. There exists a significant correlations between the histomorphometic parameters and BMD,and biomechanical parameters.
