背景:海马组织一氧化氮含量和一氧化氮合酶(nitricoxidesynthase,NOS)活性异常可能是肥胖发病的重要因素之一,已知针灸减肥机制涉及神经及神经体液调节,针刺对海马组织一氧化氮含量和NOS活性的调整作用如何?目的:探讨针刺对肥胖大鼠海马组织一氧化氮和NOS含量的影响,进一步深化针刺减肥的中枢神经作用机制。设计:以自身对照、相互对照实验研究。单位:南京中医药大学第二临床医学院针灸研究所、南京人口管理学院生殖医学教研室。材料:本实验在南京中医药大学第二临床医学院针灸研究所完成。1月龄刚断乳SD雄性大鼠,体质量50~70g,由南京军区总医院实验动物中心提供犤许可证号:SYXK(苏)2002-0017犦,动物级别为清洁级(合格证号:苏动质字第97001号)。干预:以普通全价鼠饲料喂养的大鼠作为正常组,将造模成功的实验性肥胖大鼠随机分为对照组和针刺组,每组6只。针刺组大鼠给予针刺治疗14d,正常组和对照组大鼠分别每天放入大鼠固定器中适应15min,持续14d。采用神经生化技术观察针刺治疗前后肥胖大鼠体质量、体脂及海马组织一氧化氮和NOS含量的变化。主要观察指标:①针刺对实验性肥胖大鼠肥胖指标及对心包、肾周和附睾脂肪量的影响。②针刺对各组大鼠海马组织一氧化氮含量和一氧化氮合酶活性的影响。③大鼠海马组?
BACKGROUND:The abnormality of the content of nitrogen monoxide(NO) and activity of nitrogen monoxide synthase(NOS) may be an important factor of obesity.It has been known that the mechanisms of acupuncture treating obesity are related to nervous and neurohumoral regulation.What is the regulating effect of acupuncture on NO content and NOS activity in hippocampal tissues? OBJECTIVE:To study the effects of acupuncture on the content of NO and NOS,and further investigate central nervous functional mechanism of reducing weight by acupuncture. DESIGN:Self-controlled and mutual controlled experiment.SETTING:Acupuncture Institute of Second Clinical Medical College,Nanjing University of Traditional Chinese Medicine,and Department of Reproduction Medicine,Nanjing College for Population Programmed Management. PARTICIPANTS:This experiment was carried out in the Acupuncture Institute of Second Clinical Medical College.One-month old male SD rats just in ablactation,weighing 50-70 g were provided by the Animal Experiment Center of General Hospital of Nanjing Military Area Command[Certification No.SYXK(Su) 2002-0017], and the animals were in the clean grade(Certification No. Su Animal Quality 970101). INTERVENTIONS:Rats fed with ordinary diet were in the normal group. Experimental obese rats models were randomly divided as control group and acupuncture group,6 in each.Rats in the acupuncture group were given acupuncture treatment for 14 days, and rats in the normal and control groups were put into rat fixation-machine for 15 minutes,lasting for 14 days.Body mass,body lipid and contents of NO and NOS in hippocampal tissues in obese rats before and after acupuncture were observed with nervous biochemical technology. MAIN OUTCOME MEASURES:①Effect of acupuncture on obesity index,fat contents of pericardium,kidney and epididymis of obese rats.②Effect of acupuncture on NO and NOS contents in hippocampal tissue of obese rats.③Correlation of NO content and NOS activity with body mass and Lee's index in hippocampal tissue of obese rats. RESULTS :The body mass[(458.33±7.43)g],Lee's index (302.23 ±4.11) and body fat content of obesity were all significantly higher than those in the normal rats(t=8.86,9.99,8.31,P< 0.01);NO content[(0.31±0.08) μmol/g] and NOS activity [(1.67±0.65) μkat/g] in the hippocampal tissue of obese rats were all markedly higher than those in the normal rats(t=-6.36, -4.91,P< 0.01).NO content and NOS activity were highly positively correlated with body mass and Lee's index(r=0.760, 0.804).Acupuncture significantly decreased NO content [(0.33±0.10)μmol/g] and NOS activity[(4.17±1.07)μkat/g] in the hippocampal tissue of obese rats as compared with the control group. The differences had significant meaning(t=5.24,2.88,P< 0.05-0.01).CONCLUSION:Acupuncture has fairly good regulating effect on NO content and NOS activity in hippocampal tissues of obese rats.
