粉防己碱对PDGF、TGF-β诱导瘢痕成纤维细胞增殖与胶原合成的抑制作用 被引量:8

摘要 目的 :探讨粉防己碱对血小板衍生生长因子 (PDGF BB)、转化生长因子 β(TGF β)诱导人增生性瘢痕成纤维细胞增殖和胶原合成的干预作用。方法 :体外培养的人增生性瘢痕成纤维细胞加入PDGF BB、TGF β、粉防己碱 ,用3 H TdR掺入法和3 H 脯氨酸掺入法测定粉防己碱干预下PDGF BB、TGF β诱导人增生性瘢痕成纤维细胞增殖和胶原合成作用的变化。 结果 :粉防己碱在不抑制瘢痕成纤维细胞DNA合成的药物浓度 (3 μg/ml)时即可显著抑制其胶原合成 ,并可显著抑制PDGF BB和TGF β诱导的实验细胞DNA及胶原合成。结论 :粉防己碱可抑制PDGF BB、TGF Objective: To investigate the inhibitory effect of tetrandrine on the proliferation and collagen synthesis of human scar derived fibroblasts stimulated by PDGF and TGF β. Methods: In vitro cultured fibroblasts of human hypertrophic scar were treated with PDGF BB, TGF β, tetrandrine . The effects of tetrandrine on the proliferation and collagen synthesis of human scar derived fibroblasts stimulated by PDGF and TGF β were evaluated by the incorporation method of 3H TdR and 3H Proline. Results: Significant reduction in the 3H Proline incorporation of human scar derived fibroblasts was seen at 3μg/ml tetrandrine. And also tetrandrine obviously inhibit the 3H TdR and 3H Proline incorporation of scar derived fibroblasts stimulated by PDGF BB single or together with TGF β. Conclusion: The dose of tetrandrine below the level of inhibiting fibroblast DNA synthesis could antagonize the proliferation and collagen synthesis stimulated by PDGF and TGF β in human scar derived fibroblasts. These results suggest that tetrandrine maybe unfold the potential applied prospects for the prevention and treatment of hypertrophic scars.
