摘要
目的研究活性氧(ROS)和线粒体膜电位(MMP)在高三尖杉酯碱(HHT)诱导骨髓增生异常综合征细胞MUTZ-1凋亡过程中的变化,以期进一步探讨HHT诱导细胞凋亡的机制。方法以100ng/mlHHT作用于MUTZ-1细胞一定时间后应用流式细胞仪,以AnnexinVFITC和PI双染的方法检测细胞凋亡率,ROS捕获剂HE反应法检测细胞内ROS的水平,JC-1染色法检测细胞MMP的变化。结果HHT作用MUTZ-1细胞0、6、12、24h后的凋亡率分别为5.68%±0.52%、16.88%±0.96%、43.75%±2.33%、55.48%±3.67%。在细胞凋亡的同时,细胞内ROS水平升高(P<0.01),线粒体膜电位下降(P<0.01)。结论细胞内ROS水平升高和线粒体膜电位下降是HHT诱导MUTZ-1细胞凋亡的重要机制之一。
Objective:To investigate of the changes of reactive oxygen species (ROS) and mitochondrial membrane potential (MMP) in the homoharringtonine(HHT)-induced apoptosis of MUTZ-1cell. Methods: MUTZ-1 cells were loaded with fluorescent probes Annexin VFITC, propidine iodide (PI), Hydroethidine (HE) and 5, 5', 6, 6'-tetrachloro-1,1',3,3'-tetraethylbenzimidazolcarbocyanine iodide (JC-1) , respectively .The apoptotic rate, changes of intracellular ROS and MMP were detected by flow cytometery. Results: The apoptotic rate of MUTZ-1 cells treated by HHT for0, 6, 12 and 24 h reached 5.68%±0.52%, 16.88%±0.96%, 43.75%±2.33 % and 55.48%±3.67%, respectively. The induction of apoptosis by HHT was associated with an increase in intracellular ROS(P<0.01) and a decrease in MMP (p<0.01). Conclusions: Changes of ROS and MMP may play an important role in the HHT-induced apoptosis of MUTZ-1 cells.
