TGF-β1对醛糖还原酶在大鼠系膜细胞及人肾小球肾炎中表达的影响 被引量:13

Effect of Aldose Reductase Expression in Rat Mesangial Cell and Human Glomerulonephritis by Transforming Growth Factor-β1

摘要 目的 探讨转化生长因子 β1(tranforminggrowthfactor betal,TGF β1)对其反应性基因产物醛糖还原酶(aldosereductase ,AR)在大鼠系膜细胞中表达的影响及两者在人肾小球肾炎中的表达及其相互关系。方法 采用RT PCR、Westernblot法观察外源性TGF β1对大鼠系膜细胞中AR表达的影响 ;以 12例肾小球轻微病变作为对照 ,对各种不同组织学类型的IgA肾病 (15例 ) ,膜性肾炎 (14例 ) ,局灶节段增生性肾炎 (12例 ) ,狼疮性肾炎 (14例 ) ,新月体性肾炎 (9例 )的人肾穿刺活检标本 ,应用免疫组织化学ABC方法分别观察TGF β1和AR在肾炎组织中的表达 ,并对其染色强度作图像定量分析。结果 外源性TGF β1作用后 ,大鼠系膜细胞中AR的mRNA表达与蛋白水平升高 ,并显示出一定的时间与浓度依赖性。各型肾炎中TGF β1的表达均显著高于对照组 ;除IgA肾病之外的各型肾炎中AR的表达均显著高于对照组 ,AR的表达与TGF β1表达之间具有显著的相关性。结论 TGF β1相关反应性基因产物AR的表达与TGF β1的作用密切相关 。 Purpose: To explore the effect of transforming growth factor-beta1 (TGF-β1) on its responsive gene, aldose reductase expression in rat mesangial cell and human glomerulonephritis and their significance in the development of glomerulosclerosis. Methods: Using RT-PCR, Western blot to detect the effect of aldose reductase expression in rat mesangial cell by TGF-β1. Using immunohistochemistry to dectect the expression of TGF-β1 and aldose reductase in renal biopsies from 76 cases with a spectrum including IgA nephropathy (15 cases), membranous nephritis (14 cases), lupus nephritis (12 cases), focal segmental nephritis (12 cases) and crescent nephritis (9 cases), compare with 12 cases of glomerular mild lesion with image analysis system. Results: Elevated levels of aldose reductase mRNA and protein were found in culture rat mesangial cells with stimulation of TGF-β1, and the elevation shows dose and time dependency of TGF-β1. All other types of glomerulonephritis showed increased expression of TGF-β1 and aldose reductase (AR) expcept IgA nephropathy when compared the 12 cases of glomerular mild lesion, the correlation of the expression of TGF-β1 and AR is positive. Conclusions: As one of its responsive genes, the expression of aldsode reductase in mRNA and protein level was closely related to the effect of TGF-β1, may be involved in the pathology of glomerulonephritis.
