文蛤、青蛤和四角蛤蜊的随机扩增多态性DNA(RAPD)的比较分析 被引量:1

摘要 利用随机扩增多态性 DNA (RAPD) 技术,对吕四海区的三种重要的水产贝类:文蛤、青蛤和四角蛤蜊的遗传多样性进行了分析和比较。用 60 个随机引物对三种贝类共 24 个样品进行随机扩增,从中选取了 45 个扩增效果比较稳定的引物用于群体分析,共得到了 250 个位点。根据 Nei 氏公式计算出 3 种贝类种间和种内的遗传相似性指数和相对遗传距离,并进行聚类分析。通过比较分析,3 种贝类之间的遗传距离差异不大;相对与青蛤和四角蛤蜊,文蛤种内的遗传距离要小。 Meretrix meretrix, Cyclina sinesis and Mactra vecerifermis are important aquaculture shellfish in Jiangsu Province. The genetic polymorphisms of these shellfishes, collected from the Lusi coastal area, were studied by the randomly amplified polymorphic DNA technique. Sixty random primers were used to test the 24 samples of three kinds of shellfishes. Forty-five primers were chosen for generating a total of 250 highly steady bands. According to Nei's formula, the genetic comparability index was counted. A phylogenetic tree was constructed using the UPGMA program based on the indexes of three species of shellfishes. The quantities of Meretrix meretrix's germ plasm have been declining through studies of comparing genetic polymorphisms of these three shellfishes.
