参麦注射液对缺氧/缺糖/复氧诱导神经细胞损伤的影响 被引量:3

Effect of Shenmai Injection on the Injured Neurons Cell Following Hypoxia/Hypoglycemia/ Reoxygenation

摘要 目的 探讨参麦注射液对缺氧/缺糖/复氧诱导神经细胞凋亡及胞浆内游离钙的影响,阐明参麦注射液治疗缺血性中风急性期的脑保护作用机制。方法 用孕 17~18 d 的 SD 胎鼠大脑皮层作原代神经细胞培养,制作缺氧/缺糖/复氧致神经细胞损伤模型,采用 AnnexinV/PI 双染色法以流式细胞仪定量分析神经细胞凋亡率;FIuo-3 负载,流式细胞仪定量分析胞浆内游离钙的变化。结果 参麦注射液能降低缺氧/缺糖/复氧原代培养神经细胞的凋亡率和胞浆内游离钙浓度。结论 参麦注射液治疗急性缺血性中风的疗效机制与提高神经细胞耐缺血缺氧能力,抑制细胞内钙超载,从而减轻凋亡有关。 Objective To investigate the effect of Shenmai injection (SMI) on the injured neurons cell following hypoxia/hypoglycemia/reoxygenation and to clanify the intrinsic mechanism of SMI treating acute ischemic apoplexy and protecting brain. Methods The cortical neurons of E 17~18 days fetal rat was primarily cultured. The injured model of primary cultured cortical neurons following hypoxia/hypoglycemia/reoxygenation was established. Effect of SMI on cytosolic free calcium concentration as well as the rate of apoptosis following hypoxia/hypoglycemia/reoxygenation was analyzed quantitatively by flow cytometry. Results SMI could reduce the rate of apoptosis and lower the concentration of cytosolic free calcium remarkably. Conclusion The intrinsic mechanism of SMI treating acute ischemic apoplexy has relation to the fact that SMI can inhibit the calcium overload and apoptosis of neurons induced by hypoxia/hypoglycemia/reoxygenation.
