摘要
目的探讨盾叶薯蓣组织培养的最佳外植体和激素组合。方法采用盾叶薯蓣茎段、叶为外植体 ,接种于不同激素组合的培养基上 ,观察比较培养情况。结果最佳外植体为腋芽萌发而来的无菌枝条切段 ,诱导愈伤组织的最佳培养基为MS +BA 1 .5mg·L-1+NAA 0 .5mg·L-1,不定芽分化的培养基为LS +BA1 .5mg·L-1,诱导生根的培养基为 1 2MS+NAA 1 .0mg·L-1。
Objective To determine the optimum combining conditions through investigating the effects of different combination with explants and hormones on the cultured tissues of Dioscorea zingiberensis C.H.Wright.Method Tissue cultures were made with blade and stem of this plant as explants cultured with different combination of hormones.Then, the growing status of tissues was observed and compared among diffexrent combinations. Results It was shown that the optimum explant was aseptic twig sections of auxillary bud germinating, while the optimum prescription of medium for inducing callus was composed of MS+BA 1.5?mg·L -1+NAA 0.5?mg·L -1,that for inducing abventitous buds differentiating was composed of LS+BA 1.5?mg·L -1 and that for inducing roots growing was composed of 1/2 MS+NAA 1.0?mg·L -1.Conclusion The selection of explant is the key step for culturing the tissue of Dioscorea zingiberensis successfully.
