摘要
目的 研究心肌梗死后心力衰竭(心衰)对大鼠的空间学习和记忆能力的影响。方法 采用结扎左冠状动脉前降支的心肌梗死模型,以无创性阻抗法测定血流动力学指标,利用Morris水迷宫学习记忆的实验方法,观察大鼠在心肌梗死心衰后10d、30d、60d和终末期的学习记忆能力。结果 (1)术后各个时期大鼠均出现不同程度的心衰表现。(2)心衰10d组大鼠Morris学习记忆稳定期(后8组次训练)平均对数潜伏期,手术组为(0.971±0.467)s(n=128),假手术组为(0.819±0.355)s(n=112),手术组明显比假手术组长;搜索策略手术组比假手术组明显差(x2=8.142,P<0.005);心衰30d时没有明显变化;心衰60d时大鼠的对数潜伏期没有明显变化,但搜索策略手术组比假手术组明显差(x2=4.988,P<0.05);终末期组大鼠(9组次训练)的平均对数潜伏期,终末期为(1.606±0.479)s(n=80),终末对照为(1.165±0.469)s(n=80),终末期明显比对照组长;搜索策略,终末期比对照组差(x2=11.82,P<0.005)。结论心肌梗死心衰后大鼠的空间学习记忆能力,在10d时明显受损,在30d时恢复正常,到60d时开始出现一定程度的认知损伤,在心衰终末期时认知功能明显变差。
Objective To investigate effect of congestive heart failure (CHF) on learning and memory a-bilities in the rats after acute myocardial infarction. Methods The rats underwent thoracotomy and left front descending coronary artery ligation while sham ligation control rats underwent thoracotomy without ligation. The learning and memory abilities of both ligation rats and sham ligation control rats were evaluated 10 days(n =8, n = 7) , 30 days(n= 10, n =8) , 60 days(n =9,n = 10) latter after ligation and the end-stage CHF ( n =7, n = 7) by using Morris water maze(MWM) test. The cardiac functions of all rats were examined by using the impedance cardiac output system and their cognitive abilities were tested by MWM test. Results 10 days later after ligation, the average logarithmic escape latency for the last 8 trials of ligation rats was significantly longer than that of sham ligation control rats, denary logarithm (0.971±0.467 ) s versus denary logarithm (0.819±0.355 )s, and the percents of total tendency or straight swim tracks of ligation rats were lower than those of the sham control rats, 75. 0% and 89. 3% respectively. At the end-stage of CHF after ligation, the average logarithmic escape latency for all 9 trials of the end-age CHF rats was significantly longer than that of ligation control rats which would not be die soon, denary logarithm(1. 606±0. 479) s versus denary logarithm(1. 165±0.469) s. The swim tracks of end-stage CHF rats were mainly along the walls of the pool, and the percents of total tendency or straight swim tracks of the ligation rats with end-stage CHF were lower than those of the common ligation control rats, 18. 6% and 45.7% respectively. Conclusion The learning and memory abilities of the rats with CHF significantly decreased 10 days after ligation, then turned to normal 30 days after ligation,showed a certain decline 60 days later after ligation, and decreased again at the end-stage of CHF.
