摘要
目的:观察肺癌组织中MDM2、P53蛋白的表达及其相互关系。方法:用Westernblot方法,检测 31例肺癌组织及相应 21例癌旁 3cm组织和 12例癌旁 5cm组织中MDM2、P53蛋白的表达。结果:①肺癌组织中MDM2、P53表达量高于癌旁组织。②肺癌组织中MDM2与P53表达量之间呈正相关 (r=0. 432, P<0. 05)。③肺鳞癌组织中MDM2、P53蛋白增高表达率分别为 77. 3% (17 /22 )和 72. 7% ( 16 /22 );肺腺癌组织中 2者增高表达率均为100% (7 /7), 1例小细胞肺癌和 1例大细胞肺癌组织中MDM2蛋白表达均降低,而P53蛋白表达均增高。结论:MDM2过量表达可能是肺癌发生的一个重要标志,在肺癌的发生发展过程中可能有重要作用。
Aim: To detect the expressions of MDM2 and P53 in lung cancer tissue and analyze the relationship of protein expression between MDM2 and P53. Methods: Western blot was used to detect the expression of MDM2 and P53 in 31 cases of lung cancer tissue and 21 cases of adjacent tissue. Results: The expressions of MDM2 and P53 protein in lung cancer tissue were significantly higher than those in adjacent tissue. In 31 cases of lung cancer tissue, the expression of MDM2 protein had positive correlation with the expression of P53 protein (r=0.432, P<0.05). In lung squamous cancer tissue, 77.3%(17/22) cases had overexpression of MDM2 protein and 72.7%(16/22) cases had overexpression of P53 protein.Seven cases of lung adenocarcinoma had overexpression of both MDM2 and P53 proteins, and 1 case with small cell lung cancer and 1 case with big cell lung cancer had lower expression of MDM2 protein and overexpression of P53 protein. Conclusion: The overexpression of MDM2 protein in lung cancer tissue may be an important marker and play an important role in the development of lung cancer
