摘要
采用HPLC法,以白藜芦醇的含量为指标,通过单因素实验及正交实验方法确定了从虎杖中提取白藜芦醇的最佳工艺条件。HPLC法的色谱分析条件为:YWG ODS色谱柱(长×直径为250mm×4.6mm,粒径为5μm),以φ甲醇∶φ水=45∶55为流动相,流速为0.5mL/min,检测波长为303nm。HPLC法测定结果表明,以体积分数为80%的乙醇水溶液为提取剂,液固比为14∶1,在50℃下提取2次,每次提取2h,从虎杖中提取的白藜芦醇质量分数可达0.301%。
The extract conditions of resveratrol from Polygonum Cuspidatum Sieb et Zucc were investigated by using the orthogonal design with resveratrol as index,which was determined by HPLC.HPLC detemination conditions were:YWG column(250 mm×4.6 mm, diameter 5 μm), with the mobile phase of 45% CH_3OH(ratio of volume), the flow rate of 0.5 mL/min and the detection wave length of 303 nm.The results show that the optimum extract conditions of resveratrol from Polygonum Cuspidatum Sieb et Zucc are that, with 14 times amount of (80%)(ratio of volume) alcohol under 50 ℃, the extraction can be done twice, two hours each time.HPLC determination shows that under this extract conditions ,the content of resveratrol in Polygonum Cuspidatum Sieb et Zucc is 0.301 % approximately.
