摘要
采用单因素和正交试验对纳豆菌摇瓶发酵生产纳豆激酶的工艺条件进行了优化,确定了摇瓶发酵培养基最佳配方:玉米粉0.5%,豆渣2%,麸皮0.5%,接种量为 4%,初始 pH值自然,培养温度 35℃。采用最适培养基和优化工艺,在250 ml锥形瓶中进行验证实验,纳豆激酶的酶活可达到714.2 IU/ml。
The technology of liquid fermentation for producing Natto kinase were studied by using single factor research and orthogonal test.The results showed that the optimal culture conditions were as follows:corn starch 0.5%,soybean residue 2%,wheat bran 0.5%; the inoculum size 4%; temperature 35℃.Natto kinase activity was 714.2 IU/ml when the strain was grown on the optimal medium under the above culture conditions in 250 ml Erlenmeyer fiask.
