摘要
用高效毛细管电泳法对尼可地尔及其分解物〔N (2 羟乙基)烟酰胺〕进行了分离和定量测定研究。 以烟酰胺作内标,在电压23kV、波长264±2nm、pH5.3的磷酸二氢钾 磷酸氢二钠缓冲体系中,尼可地尔与 分解物可达基线分离。尼可地尔和分解物浓度分别在12.0~264.18mg/L和2.10~43.80mg/L范围内与峰 面积呈良好的线性关系。
A method for the determination of nicorandil (NIC) and it's decomposed product N-(2-hydroxyethyl) niacinamide(HN) by high-performance capillary electrophoresis is described. The optimal separation was carried out at 22 +/- 1 degrees C 23 kV, detection wavelength 264 +/- 2 nm, using niacinamide as internal standard and 25 mmol/L KH2PO4 -Na2HPO4 buffer at pH = 5.3. There is a good linear relationship between the peak areas and the concentration of the analytes in the range of 12.0 similar to 264.18 mg/L for NIC and 2.10 similar to 43.80 mg/L for HN. The proposed method is simple, quick and accurate. It can be used to control the quality of NIC in production process, to investigate the remained samples in clinical studies.
