摘要
从皖北砀山盐碱地中分离到一株耐盐细菌 ,该菌在肉汤培养基上[1] 可耐盐至 7% ,在基本培养基上可耐至 3% .革兰氏阳性杆菌 ,具芽孢 ,芽孢表面有明显纵状突起 ,孢囊膨大 ,周生鞭毛 ,接触酶阳性 ,65℃不生长 ,可以葡萄糖为底物产酸产气 ,G +C含量为 52 .7mol% ,根据以上特性 ,检索鉴定该菌为浸麻芽孢杆菌 (B .macerans )并对其质粒进行了提取和消除实验 ,证明质粒大小约为13Kb ,耐盐性状与质粒关系不大 .同时对菌体裂解物作了SDS -PAGE凝胶电泳 ,证实在高盐( 5% )情况下比低盐 ( 0 .5% )情况下的电泳图谱多出一条蛋白质带 .此外还研究了耐盐性与培养基成分之间的关系 .
A halotolerant bacteria was isolated from the salt soda soil in the northern part of Anhui Province. The bacteria can grow well in Broth medium containing 0.5%-7% NaCl and in minimal medium containing 0.5%-3% NaCl. It is a Gram postive bacteria. In the surface of its spore, there are apparent vertical spine shaped structure. It also can utilize gluose to produce acid and gas, The G+C content is 52.7mol%. The Strain is identified as B.macerans. The organism contained a 13kb plasmid. Meanwhile we analysed the intracelleular protein under different growing conditions by SDS-polyacrylamide gel electrophoresis.This paper also explores the relation of salt tolerance and the composition of the medium.
