大豆磷脂酰胆碱提高幼年大鼠学习记忆能力及对海马CA3区突触可塑性的影响 被引量:4

Effect of phosphatidylcholine from soybean lecithin on the improvement of learning and memory of young rats and the synaptic plasticity in the region of hippocampal CA3

摘要 目的:观察大豆磷脂酰胆碱(phosphatidylcholinefromsoybeanlecithin,SB-pc)对幼年大鼠空间辨别性学习记忆能力及海马CA3区突触可塑性的影响,探讨海马突触可塑性机制及SB-pc作用机制。方法:5周龄雄性Wistar大鼠33只,随机分为SB-pc添加喂养组(11只),迷宫对照组(11只),正常对照组(11只)。喂养8周,SB-pc添加喂养组和迷宫对照组大鼠在Morris水迷宫中进行连续4d的空间辨别性学习记忆能力检测,计算机分析系统自动记录逃避潜伏期(escapelatency,EL)等相关参数,分析大鼠行为变化。用免疫组织化学方法检测SB-pc添加喂养组、迷宫对照组和正常对照组3组大鼠海马结构CA3区突触素(synaptophysin,SYN)的免疫表达。用电镜观察海马CA3区突触结构的变化。结果:SB-pc添加喂养组4d的EL分别小于迷宫对照组,差异有显著性意义(P<0.05)。SB-pc添加喂养组海马结构CA3区SYN免疫反应阳性产物的吸光度(0.5400±0.0245)高于迷宫对照组(0.4679±0.0247),差异有显著性意义(P<0.05);迷宫对照组吸光度大于正常对照组(0.3581±0.0133),差异有显著性意义(P<0.05)。SB-pc添加喂养组海马CA3区突触数密度犤(104.51±9.68)个/μm3犦和突触活性区膜面积犤(0.067±0.009)μm2/μm3犦分别大于迷宫对照组犤(84.97±6.53)个/μm3,(0.047±0.005)μm2/ AIM:To observe the effects of phosphatidylcholine from soybean lecithin(SB pc) on the spatial discrimination of learning and memory of young rats and the synaptic plasticity in the region of hippocampal CA3,and investigate the mechanism of hippocampal synaptic plasticity and the effective mechanism of SB pc.METHODS:A total of 33 male Wistar rats with 5 weeks old were randomly divided into SB pc additive feeding group,maze control group and normal control group with 11 rats in each group.After 8 week feeding,the spatial discrimination of learning and memory of rats in SB pc additive feeding group and maze control group were tested with Morris water maze for 4 days.The related paramete such as escape latency(EL) was recorded automaticly by computer in order to analyze the changes of rats' behavior.The immune expression of synaptophysin(SYN) in the region of hippocampal CA3 were tested with immunohistochemical method in SB pc additive feeding group,maze control group and normal control group.Moreover, the changes of SYN in the region of hippocampal CA3 were observed with electron microscope.RESULTS:After four days, the EL of SB pc additive feeding group was shorter than that of maze control group,which was significant difference(P< 0.05). The absorbency(A) of positive product of SYN immunoreaction in the region of hippocampal CA3 was (0.540 0±0.024 5) in the SB pc additive feeding group,which was higher than that(0.467 9±0.024 7) in maze control group, and this was significant difference(P< 0.05). A in maze control group was higher than that(0.358 1±0.013 3) in normal control group,which was significant difference(P< 0.05). In SB pc additive feeding group, the density of SYN and the area of SYN membrane in the region of hippocampal CA3 were (104.51±9.68) /μm3 and (0.067±0.009) μm2/μm3 respectively,which were higher than those[(84.97±6.53) /μm3 and (0.047±0.005) μm2/μm3]respectively in maze control group, and there was significant difference(P< 0.05).The sity of SYN and the area of SYN membrane in maze control group were also higher than those[(29.48±2.81) /μm3 and (0.020±0.001) μm2/μm3]respectively in normal control group, and there was significant difference(P< 0.05).CONCLUSION:Additive SB pc,fed to young rats, can improve the growth of SNY and the increasing of SNY membrane in the region of hippocampal CA3,enhance the immune expression of SYN,and improve the synaptic plasticity,so as to improve the spatial discrimination of learning and memory in young rats.
