摘要
利用NaCl梯度洗脱DEAE-Sepharose cL-6B柱层析纯化重组人白细胞介素1β(rhlL-1β)。rhlL-1β成分在100mmol/L NaCl浓度处被洗脱,经SDS-聚丙烯酰胺凝胶电泳鉴定为一条带,分子量约17.5KD。以此产品作为抗原,可制备出具有良好特异性的抗IL-1β单克隆抗体。
Recombinant human interleukin 1β was purified by DEAE-Sepharose CL-6B column chromatography and NaCl gradient elution. When the concentration of NaCl reached 100mmol/L, rhIL-1β was eluted and detected by SDS-PAGE. The result showed a single band with a molecular weight of about 17500 Dal. The rhlL-1β was used as an antigen to prepare anti-IL-1β McAb with good specificity.
