摘要
角叉菜胶(Carrageenin,Car)致大鼠胸膜炎后8h,胸膜腔渗出液中中性白细胞游离钙浓度(Neu-[Ca(2+)]i)升高,中性白细胞钙调素活性(Neu-CaMA)增强;粉防己碱(Tetrandrine,Tet)(10~80mg·kg(-1))于致炎前30min和致炎后4h给大鼠灌胃可明显减少炎症胸膜腔渗出液量、蛋白渗出量和白细胞游出数,同时能降低Neu-[Ca(2+)]i和Neu-CaMA;在体外,Ca(2+)(32μmol·L(-1))能不同程度的对抗Tet(20~160μmol·L(-1))抑制Neu-CaMA的作用。提示:Tet具有良好的抗炎作用,其机理与抑制Ca(2+)-CaM系统有关,Tet抑制Neu-CaMA可能与其降低Neu-[Ca(2+)]i有关。
There was an increase of cytosolic free calcium concentration [Neu-Ca(2+)]i and calmodulin activity(Neu-CaMA)of the neutrophils in the pleural exudate at 8 h after carrageenin-induced pleuritis in rats. When tetrandrine(Tet) was intragastrically administered in the dosages of 10, 20, 40 and 80 mg.kg(-1) to rats at 30 min prior to and 4 h after the infliction of pleuritis, the amount, protein content and neutrophil count of the pleural exudate could significantly reduced,[Neu-Ca(2+)]i was decreased and Neu-CaMA inhibited. In addition, it was found that calcium was able to reverse the inhibition of tetrandrine on Neu-CaMA in vitro.On the basis of these findings, it is believed that the excellent anti-inflammatory effect of Tet may be related to its antagonism to calcium-calmondulin system and the inhibition of Tet on Neu-CaMA is dependent upon the decrease of [Neu-Ca(2+)]i level.
