摘要
以0.1mol/LNaOH为溶媒,18种组成蛋白氨基酸在280~310nm波长,仅色氨酸和酪氨酸有吸收峰。利用酪氨酸在两特定波长处有等吸光度值来消除复合氨基酸中酪氨酸对色氨酸的干扰,导出两特定波长处氨基酸吸光度差值△A与色氨酸浓度G_(Trp)的回归方程:△A=-0.0034973+3.8048C_(Trp),相关系数r=0.9999,测得平均回收率为99.8%,变异系数CV=1.00%
There were only absorbing peaks of tryptophan and tyrosine in 2 80~310 nm wavelengthsin 18 amino acids with 0.1 mol/LNaOH as the solvent. The tryptophan was measured in twowavelengths of equal tyrosine ABS(λ_1=300 nm,λ_2=284.5 nm) with eliminating interferenceof tyrosine. Regression equation :△A=-0. 003 497 3 + 3. 804 8 C_(Trp). Relative coefficient e-quated 0. 999 9(r=0.999 9).Average rate of recovery was 99.8%。Variation coefficient was1.00%(CV=1.00%)。
