摘要
本实验根据酶联免疫原理,利用三种酶标凝集素:HRP—WGA、HRP—COnA、HRP—SBA,对小鼠精子表面三种相应受体进行标记,从而对三种受体的数量及其所在的部位,从形态学上进行观察,发现随着精子在附睾里的成熟及获能后随时间的变化而变化。还用生物学方法分离子精子膜蛋白,经电泳转移后,再用凝集素标记糖蛋白。观察到:(1)小鼠精子表面WGA受体主要分布于顶体区和尾部,获能后顶体后区出现了WGA受体;(2)SBA受体主要分布于精子头部,随着精子从附睾头到附睾尾的转运,受体的数量有所增加,获能后这种增加很明显,且与时间呈正相关;(3)COnA受体分布比较均匀,获能后顶体后区受体数量增多,而且这种增多也与时间成正比;(4)经电泳后测定的结果与上述符合。WGA受体的分子量为43000。
According to the principle of enzyme-immunoassay(EIA),using three sort of lectins:HRP-WGA,HRP-ConA,HRP-sba three appropriate receptor in surface of mouse sperm were signed in this experiment,and then the quantity and the location of three receptors were observed by mophology.It discovered that was changing unceasingly with ripeness of sperm in epididymis and with the time after capacitation. The membrane protein of sperm was separated using the method of bichemistry,then after electrophoresis shifting,the glucoprotein was signed using lectin.It observed: 1)WGA receptor mainly distributed in spical body and tail,after capacitation, posterior spical body appeared WGA receptor;2)SBA receptor mainly distributed in head of sperm,along with transference of sperm from head to tail of epididymis,the quantity of receptor can increase,after capacitation,the increase was vary significant and it had a positive relation with time;3)distribution or ConA receptor was well-distributed,after capacitation,the ouantity of receptor in posterior spical body increased,and this increase also had a positive relation with time;4)after electrophoresis,the tested result was similar with above-mentioned.The molecular weight of WAG reveptor is 43,000.
