摘要
目的 :研究单侧输尿管梗阻导致肾间质纤维化的发生机理及肾炎防衰液对其治疗作用的机理。方法 :采用单侧输尿管梗阻 (UUO)模型 ,术后用肾炎防衰液治疗 4周 ,然后观察大鼠肾组织的病理改变 ,应用HE ,Mallory染色观察肾组织的生理病理以及肾间质胶原的生成 ,用免疫组织化学方法 ,检测转化生长因子 β1 (TGF -β1 )和α -平滑肌肌动蛋白 (α -SMA)的在间质表达 ,以探讨纤维化的发生机理和药物的作用机理。结果模型组肾间质胶原、TGF -β1 和α -SMA的表达增加 ,阳性药物苯那普利组和肾炎防衰液治疗组与模型组相比有显著差异 (P <0.0 5 ) ,表现为胶原、TGF -β1 、α -SMA的表达下调。结论 :肾炎防衰液可能通过抑制转化生长因子β1 的生成减缓了胶原的过度表达 ,延缓了纤维化的进程 。
Purpose:To observe the mechanism of prevention and cure of Shen Yan Fang Shuai Ye on rat after unilateral ureteral obstruction(UUO).Methods: SD rats were divided into five groups, after UUO,NO.4 and NO.5 groups were treated with benazepril and Shen Yan Fang Shuai Ye group by gavage for 4 weeks.After four weeks, killed the rats and observed the kidney tissue after HE and Mallory stain, Immunohistochemistry was applied to measure α-SMA TGF-βin the renal interstitium.Results: Compared with the model group, the benazepril and Shen Yan Fang Shuai Ye group had less production ofα-SMA and TGF-β.Conclusion: we can conclude that Shen Yan Fang Shuai Ye could improve the condition of the renal failure rats after UUO, and had the similar effect to the control group.
