摘要
对6只10月龄西农莎能奶山羊公羊按每日10g/(kg·d)的剂量单笼饲喂甘肃棘豆粉,18d后陆续出现中毒症状,精子呈现不同程度的损害:密度下降(最低达12亿/mL),畸形率高达29%;电镜负染色法观察其超微结构,发现精子顶体膨胀、半脱或全脱,尾梢松散;中毒精子顶体完整率(27.5%)明显低于正常精子(71.75%);用电镜超薄切片法观察精子的超微结构表明,细胞核、顶体及尾部中段线粒体空泡变性,细胞膜、顶体外膜及中段线粒体鞘扩张或断裂。
Six 10-month Xinong saanen dairy billy goats were individually fed with10g/kg of dried and ground Oxytropis Kansuensis per da After the 18th day, the billygoats appeared to have toxic symptoms in succession:and their sperms showedsomedamage in different degrees. Sperm density decresaed (the minimun density was 1.2b./mL).The percentage of abnormal sperms reached as high as 29%. The electron mi-croscopic examination of ultrastructure of sperms was carried out using negative stainingtechnique showed the changes in morphology of acrosom, being swollen, partially de-tached and detached and loose sperm tail end. The percentage of acrosomal integrity oftoxic sperms (27.5%)was significantly lower than that of normal sperms in the control(71. 75%).The electron microscopic examination of ultrastructure of sperm was carriedout using ultramicrotomy showed the vacuolation of cell nuclei,acrosome and mediummitochondria of tail end. dilation and break of cell membrane,acrosome outside mem-brane and medium mitochondria sheath.
