摘要
建立一种简单、快速的反相高效液相色谱(RP-HPLC)法测定穿心莲、路边青配伍药液中脱水穿心莲内酯在小鼠血浆中的浓度.使用C18反相柱(4.0mm×250mm,5μm),流动相为甲醇-乙腈-水(13∶17∶70),流速:1.0mL·min-1,检测波长:252nm.血药浓度在0.102~20.305mg·L-1(r=0.9998)时线性关系良好;低、中、高浓度下的回收率、重现性均符合方法学要求.该方法能有效地监测小鼠血浆中脱水穿心莲内酯的浓度变化.
To develop a simple and rapid reversed-phase high performance liquid chromatographic method for the determination of dehydroandrographolide in Andrographis and Clerodendron compatible preparation in mice plasma.The C_(18) reversed-phase column(4.0mm×250mm,5μm)was used as analytical column;The mobile phase consisted of methanol-acetonitrile-water (13∶17∶70) at a flow rate of 1.0mL·min^(-1);The detection wavelength was 252 nm. The calibration curve in plasma was linear well over the range of 0.102~20.305mg·L^(-1)(r=(0.9998));The recovery and reproducibility of low、middle and high concentration are accord with methodological request. The method is proved suitable for monitoring the change of concentration of dehydroandrographolide in mice plasma.
