摘要 目的 探讨中波紫外线 (UVB)对人角质形成细胞 (HKC)分泌白介素 - 8(IL - 8)的影响。方法 采用不同强度的 U VB照射体外培养的 HKC,2 4 h后用 EL ISA法检测上清液中 IL - 8的质量浓度 ;以及在相同强度UVB照射后的不同时间点检测 HKC分泌 IL - 8的水平。结果  U VB照射可使 HKC分泌 IL - 8增加 ,该效应在 10~4 0 m J/ cm2 时呈强度依赖性 ,UVB强度为 2 0~ 70 m J/ cm2 时各组 IL - 8质量浓度与对照组 (0 m J/ cm2 )比较差异有显著性 (P<0 .0 1)。 HKC接受 30 m J/ cm2 U VB照射后 ,其 IL - 8的分泌在 1h内开始增加 ,12 h达到高峰 ;照射后 3h、6 h、12 h、2 4 h时 HKC分泌 IL - 8的水平与 0 h时比较差异有显著性 (P<0 .0 1)。结论  U VB可使 HKC IL - 8分泌增加 ,该效应在一定范围内呈强度依赖性。 Objective To investigate the effect of ultraviolet B (UVB) on Interleukin-8 secretion in human keratinocyte cell line. Methods The concentration of IL-8 was detected by ELISA kit 24 h after human keratinocytes(HKC) were irradiated by different doses of UVB, and the level of IL-8 was also determined at different times after the same dose of UVB irradiation. Results The secretion of IL-8 was increased after the HKC were irradiated by UVB, the effect was dose-dependent when UVB ranged from 10 to 40 mJ/cm 2, and there was statistically significant difference between the IL-8 level of UVB groups (20-70 mJ/cm 2) and the control (0 mJ/cm 2) (P<0.01). The level of IL-8 was increased 1 h after the irradiation of 30 mJ/cm 2 UVB, and it reached the peak at 12 h. There was statistically significant difference between the IL-8 levels detected at different times (3 h, 6 h, 12 h, 24 h after irradiation) and that at 0 h(P<0.01). Conclusion UVB increase the secretion of IL-8, and the effect is dose-dependent to some extent.
