摘要 目的:研究抗α1肾上腺素受体自身抗体对大鼠血管结构的影响。方法:用α1肾上腺素受体胞外第二 肽段合成肽免疫Wistar大鼠作为该抗体的模型,设自发性高血压大鼠(SHR)及正常Wistar大鼠为对照;用尾套法 检测血压和用ELISA法检测抗α1肾上腺素受体抗体滴度,用光镜和电镜观察主动脉和肠系膜第三级动脉病理变 化,并计算管壁厚度、管壁/管腔比;免疫组化方法检测主动脉壁的c jun和c fos的表达。结果:在免疫后2周,免疫 组抗α1肾上腺素受体抗体滴度开始升高,并在整个实验过程中维持在较高水平;在实验过程中,免疫组血压与对 照组无明显差异,但均显著低于自发性高血压大鼠组;免疫组主动脉管壁厚度((0.382±0.075)mm)、肠系膜三级 动脉管壁/管腔比(0.526±0.209)均显著高于正常对照组(分别为(0.297±0.009)mm,0.379±0.072);电镜结果 显示免疫组较正常对照及SHR组主动脉平滑肌线粒体增多,间质胶原纤维增多;免疫组主动脉壁c jun蛋白表达高 于正常对照组,但c fos的表达与正常对照无异。结论:抗α1肾上腺素受体自身抗体可通过增加平滑肌细胞c jun 的表达而导致血管结构的改变,可能在高血压发病中发挥重要作用。 Aim: To study the vascular structure changes induced by antibodies against the α_1-adrenoceptor. Methods: The models with the antibodies against α_1-adrenoceptor were made by immunizing Wistar rats with the second extracellular loop of α_1-adrenoceptor peptides. Spontaneous hypertension rats (SHR) and normal Wistar rats served as control. The blood pressure(BP) and antibody titers were determined using tail-cuff method and ELISA,respectively.The pathological changes of aorta and mesentery were observed under microscope.c-jun and c-fos expressions in aorta were measured by using immunohistochemistry. Results: At 2 weeks after immunity,the antibody titers in immunity group began to increase and remained a higher level through out the experiment;BP in immunity group and control group had no significant difference but lower than that in SHR group;the thickness of aorta vascular wall in immunity group was higher than that in control group((0.382±0.075) mm vs (0.297±0.009) mm),and the ratio of vascular wall thickness to lumen thickness in immunity group was higher,too.Compared with control group and SHR group,in immunity group,the mitochondria of smooth muscle cells and collagen fibril in interstitum both increased;the expression of c-jun protein was higher than that of control group.Conclusion: The antibodies against α_1-adrenoceptor could contribute to vascular structure changes in rats by increasing c-jun expression of vascular smooth muscle cells, and may play a particular role in hypertension.
