摘要
为建立快速检测四环素族抗生素残留的方法 ,采用大量接种快速培养方法 ,制备出适宜检测用的发光菌检测液 ,建立了发光细菌检测四环素族抗生素体系。结果表明 ,在该检测体系条件下 ,明亮发光杆菌与金霉素、土霉素和四环素的抑制效应作用呈良好的线性关系 ,菌液与抗生素的作用最佳体积配比为 1∶3,相互作用的适宜时间为 6 0~ 80min。该体系对四环素族抗生素的检测灵敏度可达 0 0 0 6 2 5 μg ml,比传统的杯碟法高 5倍 ,具有快速、简便的特点 ,可作为食品中抗生素残留的一种快速、灵敏的筛选检测方法。
Luminescent bacteria have been widely used to monitor water quality and assess the toxicity of substances, but hitherto no reports are available on its use in detecting the biological activity of antibiotics. A rapid cultivating method of massive inoculation was adopted to produce a type of luminescent bacteria suspension suitable for detecting tetracyclines in food samples and a testing system was established. Experiments indicated that the best volume ratio of the bacteria suspension to the tetracycline solution was 1∶3 for their interactions, and the best interaction time was approximately 60~80 minutes. Under the system conditions, the inhibiting effects of oxytetracyline, chlortetracycline and tetracycline on the Photobacterium phosphoreum presented good linear relationship. The sensitivity of this system in detecting tetracyclines can reach 0 006 25?μg/ml, which is five times higher than that of the traditional cylinder plate method. The system can be used as a rapid and convenient screen method for detecting tetracycline residues in foods.
