摘要
目的 体外实验研究三氧化二砷(As2O3)对食管癌细胞侵袭转移及血管形成的影响及探讨其作用机制。方法 MTT法观察As2O3对ECA109细胞粘附能力的影响;用Boyden Chamber膜侵袭系统观察As2 O3 对ECA109 游走和侵袭以及诱导内皮细胞形成新生血管的影响;免疫细胞化学观察As2O3对ECA109细胞TGF- β1表达的影响。结果:As2O3作用后,ECA109细胞粘贴率降低(P<0.05),游走穿膜细胞数或侵袭穿膜细胞数均明显低于对照组,诱导内皮细胞形成管腔数及长度均低于对照组(P<0.05)。As2O3能降低TGF β1的表达。结论:As2 O3 能有效抑制食管癌侵袭转移及血管形成,其作用机制可能与通过降低TGF β1的表达,抑制食管癌细胞异质粘附、运动能力及血管形成有关。
Objective To study the effect of arsenic trioxide ( As 2O 3) on the invasion metastasis and angiogenesis of esophageal car cinoma in vitro. Methods MTT was used to study the effect of adhesiveness by As 2O 3 on ECA109 cells. The Boyden Chamber membrane invasion culture sy stem was used to study invasion,metastasis and angiogenesis.Immunocytochemistry was used to observe the change of TGF-β 1. Results In the membrane invasion culture system, the numbers o f invading and migrating ECA109 cells were significantly lower in As 2O 3 group than in the control group (P<0.01), and the numbers and length of tubes were significantly lower in As 2O 3 group,too (P<0.05). The adhesion rate and TGF-β 1 expression were down regulated in As 2O 3 group (P<0.05).Conclusion As 2O 3 can inhibit the migration,invasion and angiogenesis of esophageal carcinoma cells ECA109 in vitro. The mechani sm may be down-regulation of TGF-β 1 expression.
