摘要
目的 通过形态学观察,研究胰岛素对短暂脑缺血大鼠海马CA1 区迟发神经元死亡的影响。方法 线栓法制备大鼠大脑中动脉阻塞再灌注模型,胰岛素组术前腹腔注射胰岛素和葡萄糖使血糖维持在正常范围,模型组注射生理盐水,再灌注后第1、3和7天取材,HE染色观察计数海马CA1区正常神经元。结果 再灌注后1、3、7 d模型组海马CA1区正常神经元为176.00±4.24、110.75±7.89和59.00±8.41;胰岛素组为:178.00±8.52、166.00±6.06 和92.50±9.98。结论 胰岛素可延迟局灶性短暂脑缺血后海马CA1 区迟发性神经元死亡,这可能是其发挥脑保护作用的机制之一。
Objective To investigate the effect of insulin on the expression of normal neurons in hippocampus CA1 region in rats with cerebral ischemic-reperfusion. Methods Models with intraluminal occlusion middle cerebral artery were used. Rats were given intraperitoneal injection of insulin and glucose or saline prior to operation to maintain systemic glucose level in the normal range. On 1,3 and 7 d after the reperfusion. The animals were sacrificed and samples were stained for histopathological analysis to investigate the normal neurons in hippocampus CA1 region. Results There was delayed neuronal deaths (DND). On 1,3 and 7 d after the reperfusion, the number of normal neurons in hippocampus CA1 region was 176.00±4.24, 110.75±7.89 and 59.00±8.41 in model group, and 178.00 ±8.52, 166.00±6.06 and 92.50±9.98 in insulin group, respectively. Conclusion Insulin delays and reduces DND, which could be one of the mechanisms of insulin' neuroprotective effect.
