摘要 目的:观察碱性成纤维细胞生长因子(bFGF)及其受体FGFR-1在大鼠血管平滑肌细胞(VSMCs)的表达情况。方法:建立大鼠血管平滑肌细胞原代培养并鉴定;使用原位杂交检测VSMCs中bFGF及其受体FGFR-1的mRNA表达;用免疫组织化学和WesternBlot技术鉴定bFGF蛋白表达情况。结果:通过光镜和免疫组织化学技术从细胞形态和结构上使VSMCs得到鉴定;原位杂交证实大鼠VSMCs表达bFGF及其受体FGFR-1的mRNA;免疫组织化学表明VSMCs的胞核和胞浆中有bFGF蛋白表达,WesternBlot显示bFGF有18KDa、22KDa、22.5KDa3种异构体形式。结论:大鼠血管平滑肌细胞表达bFGF及其受体FGFR-1mRNA,bFGF有多种异构体形式并分布在VSMCs的胞核和胞浆中。 Objective: To investigate the gene expression of basic fibroblast growth factor(bFGF) and its specific receptor FGFR-1 in the rat VSMCs. Methods: The construction and identification of primary culture of the rat VSMCs. bFGF mRNA and its specific receptor FGFR-1 mRNA were detected by in situ hybridization. bFGF protein was evaluated by immunocytochemistry and Western blot. Results: Morphology of VSMCs was verified routinely by phase-contrast light microscopy.VSMCs were identified by immunocytochemical staining for α-actin. The result demonstrated that rat VSMCs could express bFGF and its relevant receptor FGFR-1 in a high level. The outcome shows that the expression of bFGF protein is positive in both cytosolic and nucleus of VSMCs and there are three bFGF isoforms(18KDa?22KDa?22.5KDa )by Western Blot. Conclusion: The rat VSMCs express mRNA of both bFGF and its receptor FGFR-1. The protein of bFGF is positive in VSMCs. Its isoforms are localized in cytoplasm and nucleus respectively.
